Extraction,purification,and degradation of enzyme from a PAH-degrading strain flavobacterium sp.FCN2

The Flavobacterium sp.FCN2 has been reported to be an effective PAH-degrading strain.In this paper its active endoenzyme was extracted and partial purified through salting out method,dialysis and size exclusion chromatography,and degradation characteristics of the acnve enzyme for anthrance,phenanthren.And pyrene was also studied.Comparing with other components of active endoenzyme,which obtained from 65%ammonium sulfate salting-out has the highest PAH-degrading ability.And under existence of this part of endoenzyme,within 60 minutes,the removal rates of anthrance,phenanthren,and pyrene were 56.43%,36.45%and 45.72%,respectively.This showed that this part of endoenzyme had the rgmoval efficiency for anthracene and pyrene than phenanthrene.The optimum temperature for the activity was observed to be 32℃and the optimum pH for activity was 5-7.The further pudficafion of 65%-ammonium-sulfate-salting-out component was performed by using size exclusion chromatography (the condition of chromatography:flow rate:1mL/min;filling:Superdex G75;mobile phase:0.02mol-mL-1 monopotassium phosphate and 0.10 mol-mL-1 sodium chloride,pH=7.0,two Peaks appeared on the size exclusion chromatography elution curve,the protein which retention times were about 6-10 minutes has degradation activity towards pyrene,And the specific activity of enzyme was 20.68 U-mg-1 The molecular structures are need further determined.
