Cultivation and properties of endothelial cells on anionic gel scaffolds

1. Introduction Hydrogels are appealing materials for tissue engineering due to their structural similarity to macromolecular-based extracellular matrix in biological tissue. However, there are significant biological and physical shortages of synthetic hydrogels, such as poor cellular viability and weak mechanical strength, greatly prevent the wide application of synthetic hydrogels in tissue engineering field. In physiological condition, the role of tissue cells is important in biological activity. For examples, a continuous endothelial cell (EC) monolayer at the inner surface of blood vessel contributes to protecting procoagulant activity. Needless to say.Many tissues, such as blood vessel exist in a severe mechanical dynamic environment. Therefore, from a potentially practical view point, in order to replace the natural tissues with hydrogels, design and production of hydrogels with some critical parameters, such as high cellular viability and mechanical strength, are crucially important in the biomedical applications of hydrogels. We have strived to develop hydrogels with these critical parameters by using synthetic polymers besides natural polymers. The following topics have been investigated:1) develop adhesive protein free synthetic polymer gels as scaffolds for cell cultivation,2,3,2) control cell functions by gel properties4,and 3) creation tough hydrogels for cell cultivation5.We believe these studies would substantially promote the application of hydorgels for tissue engineering.
