Rapid prototyping a double layer polyurethane-collagen conduit and its schwann cell compatibility
A new double layer conduit combining outer synthetic polyurethane (PU) layer and inner natural collagen layer was fabricated via a double-nozzle low-temperature deposition manufacturing (DLDM) technology. The outer PU layer can provide a mechanical stable tunnel against scar tissue invasion during in vivo experiments. The inner collagen layer is able to promote Schwann cell adhesion, migration and proliferation. Microporous structures were found in both the double layers. Tight connection between the double layers was achieved through adjusting the distance between two deposited nozzles and other processing parameters. Schwann cells from rat sciatic nerves were cultured in the double layer PU-collagen conduits for one week and showed significant difference in their retention ability and viability as compared to that in pure PU conduit. The double layer polyurethane-collagen conduit had a better Schwann cell compatibility and potential for clinical peripheral nerve repair.
