Fractionation of Ganoderma lucidum polysaccharide by membrane separation and its immuno-stimulating evaluation
Raw Ganoderma lucidum polysaccharide(GLP) extracted by water were fractionated using membrane technology. The flux change during the filtration and after membrane cleaning procedure were investigated. The results showed that during the filtration of polysaccharide solution, the flux of membrane decreased quickly at beginning, then it became stable a few hours later. After washed with 0.1M sodium hydroxide for half an hour, the water flux of contaminated membrane can be recovered. The bioactivity of Ganoderma lucidum Polysaccharide(GLP) fractionations was evaluated by proliferation experiment of T, B lymph cell. GLP fractionations presented different effect in its bioactivity, The GLP fractionation with molecular weight (MW) >300,000 dalton has the highest effect to improve cell immunity. All the results showed that membrane separation provided a good method to purify or fractionate the GLPs, while for the commercial application the membrane fouling has to be solved.
