Numerical Simulation of Aggregation of cell-microbeads
The aggregation of microtissue has great influence on the viability of internal cells and the in vitro fabrication of large-scale engineered tissue. In this article, we investigated the effect of cell seeding density, microcarrier concentration and Vc on the formation of microtissue and the size of aggregates in microcarrier culture system in spinner flask. We also developed a mathematical model about microtissue formation and aggregates size according to that. The results showed that the process of aggregation had three phases: lag phase, logarithmic phase and static phase. The average aggregate size of microtissue increased with the increase of cell seeding density and microcarrier concentration and the addition of Vc. Our mathematical model can successfully simulate the microtissue formation process and the aggregates size under different operation parameters.
