Anti-breast tumor activities of CpG ODN-activated dendritic cells co-cultured with cytokine-induced killer cells in vitro and in vivo

Cocultured dendritic cells (DC) and cytokine-induced killer cells (CIK) system was established to create specific and non-specific anti-tumoral immunity. Here we show that unmethylated CpG oligodeoxynucleotide (CpG ODN) enhanced the cytotoxic activity of cocultured DC and CIK cells against tumor cells in vitro and in vivo. Cocultured DC and CIK cells treated with purified 85kDa breast tumor-associated antigen showed cytotoxicity against cultured breast tumor cells. Treatment with CpG ODN significantly increased the anti-breast tumor activity of cocultured DC and CIK cells. The levels of IL-12 and CD3 |? increased in the coculture media after CpG ODN treatment. Moreover, administration of CpG ODN treated DC and CIK cells significantly reduced the size of preformed tumor in nude mice and increased the survival rate of the mice. Our results strongly suggested the usefulness of CpG ODN as a reagent for cancer therapy using cocultured DC and CIK cells system.
