COMPARISON OF DIFFERENT ANTISERA IN ORDER TO DETECT PREGNANCY ASSOCIATED GLYCOPROTEIN (PAG) IN SWAMP BUFFALO
This study was conducted in order to test the ability of three different RIA to detect PAGs in swamp buffalo fluids. The three antisera were raised in rabbit immunized against purified bovine PAG67kDa (As497 or RIA 1), against caprine PAG55+62kDa(As706 or RIA 2) and buffalo PAG (As 859 or RIA 3). The RIA systems were taken with and without preincubation of unknown sample with antibody.In all assays, the tracer and the standard were prepared from a purified preparation of PAG 1. The sera were withdrawn from pregnant buffalo at different time of pregnancy. All the RIA systems were able to detect PAG from low concentrations. The dilutions of different samples of swamp buffalo were parallel with the standard. The aforementioned systems were shown suitable for PAG determination in swamp buffalo.
