Study of Regenerated Bud Culture from Explants of Lagerstroemia Indica Leaves

. Lagerstroemia indica L is famous for its flowers especially in spring and summer. In our research, we chose new and old leaves of L. indica as explants. Based on the MS medium, we added different concentrations of auxin such as 2,4-D, 6-BA, IAA and NAA in order to study the L. indica tissue culture technology. By researching regenerated buds of L. indica and preliminary exploration, we built the asepsis propagation system and obtained the method of inducing calli and buds differentiation. At the same time, we found the high efficient approach of regenerated bud culture by choosing L. indica leaves as explants. Our experiment adopted two methods to induce buds differentiation. One was a two-step method which induced calli at first and then transferred calli into buds differentiation media. Another was that explants can produce buds directly on the medium. The results showed that puting leaves on the MS + 6-BA 2.0 mg/L + IAA 0.2mg/L medium can induce calli and produce buds, and such direct training division is the most efficient way.
