Aquaporin 1 is Involved in the Antimigration Effect Of 20 (R) - Ginsenoside Rg3

Aim:To investigate the new mechanism of 20(R)-ginsenoside Rg3 (Rg3),one of the active compound in ginseng root,on antimigration in vitro and analyze the involvement of aquaporin 1 (AQP1,the protein facilitating water trans-membrane transportation) in the inhibition process.Method:The effect of Rg3 on human prostate cancer cell (PC3M,with a high metastatic characteristic) viability and migration was analyzed by MTT assay and wound healing assay.Immunofluorescence staining was used to characterize AQP1 in PC3M cells.AQP1 protein expression with Rg3 or proteasome inhibitor MG132 and ERK pathway activation was examined by western blotting.Additionally,RNA interference was used to investigate the AQP1 involvement of PC3M cells migration inhibition induced by Rg3.Recults:Rg3 (10-8-10-5M) remarkably inhibited PC3M cells migration(P<0.05),exerting no effect on cells viabilitics.Also,AQP1 protein expression got time-dependently and dose-dependently suppressed with Rg3 treatment (P<0.05).Furthermore,AQP1 expression suppression abolished by Rg3 may attribute to ERK pathway activation not more AQP1 degradation through ubiquitin-protcasomc pathway (P<0.05).PC3M cells migration was significantly suppressed by AQP1-siRNA (P<0.05).However,PC3M migration inhibition induced by Rg3 was not completely reversed by AQP1-siRNA (P<0.05).Conclusion:Rg3 has antimigration activity,which may depend on its inhibition of AQP1.
