Preliminary Study on Molecular Recognition of Pathogenic Vibrio Based on SELEX Techniques

Based on the designment of the 76mer ssDNA library and choice of affinity determination method, a SELEX system to select the high affinity aptamers against Vibrio alginolyticus was preliminarily established and the high affinity aptamers were selected through 5 rounds of the SELEX.The study showed that the non-specific amplification in the replication of the ssDNA library was inhibited in the end phase of the SELEX. The high affinity aptamers that would be used for the molecular recognition of the microorganism were becoming more and more with the increasing SELEX rounds, though the affinity of ssDNA library decreased in some SELEX round. The affinity of the ssDNA library increased by 80% through only 5 rounds of the SELEX.
