Study on Amplification Method of ssDNA Library in Detection of Pathogenic Vibrio by SELEX
System evolution of ligands by exponential enrichment (SELEX) was applied to select aptamers that can recognize pathogenic microorganism Vibrio alginolyticus. Amplificaition of ssDNA library is the key step in this technique. The effects of asymmetric PCR method and simplified magnetic beads (SMB) method on the amplification of ssDNA library were comparatively studied in the present paper. The results show that the asymmetric PCR method has lower efficient amplification; the later purification process of ssDNA also makes it inconvenient and hard to manipulate. SMB method has higher efficient amplification for the ssDNA library. A new improved amplification method, SMB method for ssDNA library was developed by changing the purification step into neutralization step.
