Quantum Dot-Encoded Microbeads: Synthesis and Application to Microchip Flow Cytometer for Diagnosis

A series of quantum dot(QD)-encoded polystyrene microbeads is prepared. Compared with microbeads loaded with traditional organic dyes, as-synthesized fluorescence microbeads have better photostability. These microbeads could conjugate with protein by further surface modification, so they could be applied to such fields as biochemistry, molecular biology, cell biology, immunology. A home-made microfluidic system is designed and fabricated, and coupled with flow cytometry for QD-encoded microbeads analysis. The microbeads modified with biomolecules can be used for pathogen detection. A method for detecting epidemic hemorrhagic fever and new-castle disease based on such microbeads was developed. It was shown the set-up could be a good platform for detecting multiple disease pathogens, simultaneously.
