Study on Chemiluminescence Reaction of Dichlorofluorescein-Bovine Serum Albamin by Flow-Injection Method

The chemiluminescence (CL) characteristics of bovine serum albumin (BSA) labeled with dichlorofluorescein (DCF) has been studied by using flow injection method. The experimental conditions for the DCF-BSA- ClO-cetyltrimethylammonium bromide (CTAB) CL system were optimized. Based on the relations between CL intensity and BSA concentration; a new method has been established for the quantitative determination of BSA. Under the optimum conditions, the proposed method has a linear range of 0.01 to 20.0μg/ml, with a detection limit of 0.007μg/ml for BSA (3σ). The relative standard deviation of 1.0μg/ml BSA (n=8) is 1.4%. The method was applied to the determination of BSA in milk sample with satisfactory results.
