Whole Blood PCR on a Static Chip Thermostat of ITO Substrate
In this paper, PCR amplification on a static PCR chip system with whole blood sample without any purification was demonstrated. The static PCR chip system was developed based on a substrate of indium- tin oxide glass (ITO-glass) using the conductive layer as an electro-heating element. Pulse width modulation (PWM) and fuzzy proportion integration differentiation (PID) algorithms were adopted in controlling of temperature programs of the chip. Direct blood PCR amplification conditions, such as composition of reagents and thermal programs, were first studied systematically on GeneAmp PCR system, and on-chip PCR amplification from only 0.5μL blood was successfully demonstrated using the static PCR chip system. Only thermal treatment was involved in whole process. This work demonstrates that portable PCR chip system for point of care or in-field forensic analysis is possible.
