Screening a Strain of Cellulolytic Microorganism and Studying Its Physiological and Biochemical Characteristics
The 12 strains of cellulolytic microorganism were isolated from rotten corn straw and fallen leaves in the moist ground using Congo-red cellulose plating method. One strain of cellulolytic microorganism was selected and its carboxymethyl cellulose (CMC) activity and filter paper activity (FPA) were measured under conditions of various temperature, pH value and time. Orthogonal test was used to investigate main effective factors affecting this strains enzyme activity. The strain was identified by morphological, physiological and biochemical characteristics and 16SrRNA gene sequence analysis. The optimum glucose production conditions of this strain of microorganism were confirmed by morphological test: 40℃, pH=4, 6d.
