Stereoselective Studies on Different Pharmacokinetic Behavior and Tissue Distribution of Theta-cypermethrin Enantiomers in Rabbit
Theta-cypermethrin (TCYM) is a potent pyretroid insecticide. Enantioselective kinetics, tissue distribution and chiral inversion of TCYM in rabbits were investigated via intravenous injection. The concentrations of TCYM enantiomers in samples were determined by a HPLC with Diode Array Detection (DAD) and a cellulose-tris-(3, 5-dimethylphenylcarbamate)- based chiral stationary phase (CDMPC). After intravenous administration of racemic TCYM, the (-)- (R)-(1S)-trans-TCYM was prevailing in plasma. The(-)- (R)-(1S)-trans-enantiomer of the area under the concentrationtime curve (AUC<,0-∞>)was 248.94μg/(mL· min). The total plasma clearance value (CL) of (-)-(R)-(1S)-trans- TCYM was 30.00mL/(min·kg). The (-)-(R)-(1S)-trans-TCYM was enantioenriched in tested tissues except in lung. These results indicated substantial stereoselectivity on kinetics and tissues distribution of TCYM enantiomers in rabbits.
