Application of Folate Mediated Tat and Drug -loaded Magnetic Nanoparticles in Epilepsy Cure

This paper described a new formulation of magnetic nanoparticles coated by a novel polymer matrix----O-Carboxylmethylated Chitosan (O-CMC) as drug carrier. The O-CMC magnetic nanoparticles were modicated with a peptide sequence from the HIV-Tat protein and folate (FA) to improve the translocational property and cellar uptake of nanoparticles. To evaluate the effectiveness of this drug carrier (O-CMC-MNPs-Tat-FA), sodium valproate (SVP) was incorporated as a model drug and SVP-loaded O-MNPs-Tat-FA with the average diameter of 90nm were prepared and characterized by Transmission Electron Microscopy (TEM), Dynamic Laser Light Scattering, X-ray Diffraction (XRD) and Vibrating Samples Magnetometer (VSM). We also studied the ability of SVP-O-CMC-MNPs-Tat-FA crossing blood-brain barrier (BBB) in rats by single photon emission computed tomography (SPECT). The result of curing epilepsy was investigated by in vivo test. The vivo test indicated that Tat peptide and folate helped the drug carrier to successfully pass the caption of blood brain-barrier (BBB) under the magnetic field, and as well as had certain positive effects on therapying epilepsy. Thus, the drug system had great promise for therapying disease in the brain.
