Synthesis and characterization of novel amphiphilic multifunctional octadecyl quaternized carboxymethyl chitosan
The design for the construction of more-effective delivery vectors is very important. Typically, a new kind of multifunctional octadecyl quaternized carboxymethl chitosan(QACMC) with varying degree of substitution (DS) and molecular weight (Mw) were prepared using nucleophilic substitution reaction, the structure of the modified chitosan were characterized by FT-IR,1H-NMR, XRD and gel permeation chromatography. The results showed that QACMC has many unbeatable advantages. It exhibited excellent solubility both in water and organic solvents such as chloroform, DMF, THF and toluene when Mw was above 1×104, which greatly expands its application. QACMC has amino group, carboxymethyl salt group and octadecyl quaternized group simultaneously, so targeting materials, such as antibodies, can be connected to some of these chemical groups. It also has a high and perfect crystallinity compared with chitosan and other derivatives. TEM images indicated that it can be made into spherical micelles and double-membrane structure micelles. QACMC micelles also have the solubilization function of pyrene. Zeta potential of QACMC could reach +52.72mV when grafting percentage of quaternary group was about 105.0%, which means that gene and some proteins can be adsorbed on the surface of the nanoparticles through static effect. QACMC can be considered as a good potential vector for genes and drugs.
