A DNA Electrochemical Sensor Prepared by Nano Au/p-aminothiophenol Self-assembled Films and Its Application for Label-Free PAT Gene Fragment Detection

p-Aminothiophenol (PATP)/Au self-assembled film was formed via the strong Au-S linkage. The mixture of nanogold and 3-mercaptopropionic acid (Au-MPA), and diallyldimethylammonium chloride (PDDA) were successively modified onto the PATP self-assembled film to prepare gold nanoparticle multilayer films. The performance of the gold nanoparticle multilayer films was studied by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The prepared modified electrode was used as a platform for the immobilization and hybridization of DNA, which were characterized by EIS. This DNA electrochemical biosensor was successfully applied to the detection of the PAT gene fragment by a label-free EIS method. The dynamic detection range was from 1.0×10-11 mol/L to 1.0×10-6 mol/L 20-base PAT gene fragment, with the detection limit of 2.3×10-12 mol/L.
