A DNA Electrochemical Biosensor Based on HRP and CNT
In the experiment, carbon nanotube (CNT) was treated with sonication and circumfluence to build carboxyl group on its surface, then, dropped the functionalized CNT onto the bare CPE surface to attain the CNT modified electrode. As carboxyl group and amine could covalently link together, target DNA with 5′-NH2 could be easily immobilized onto the surface of the modified CPE with the help of activation reagent EDC and NHS. CNT was also used as a bridge to label horseradish peroxidase (HRP) to probe DNA. After hybridization, electrochemical method was applied to detect the HRP on the electrode. In this way, target DNA was detected indirectly. The experiment conditions were optimized and the calibration curve was obtained. The detection range of target DNA with this biosensor was 1.0×10-11-1.0×10-7 mol/L and the detection limit was 2.5×10-12 mol/L.
