Preparation and Characterization of dual-biomolecule-labeled nanogold probe
Objective An novel gold nanoparticle probe were prepared in this paper , antibody and thiol-capped oligonucleotides combined with gold nanoparticles. Methods To form the novel nanogold probe ,the antibody and thiol-capped oligonucleotides were firmly combined with gold nanoparticle at the same time through electrostatic force and covanlt binding. Using Transmission electron microscopy(TEM)、UV -vis spectra to describe the morphology and spectroscic properties of probes; dot-immunogold filtration assay and Immunogold-silver staining to detect antibody activity; using dithiothreitol(DTT) to displace thiol-capped oligonucleotides of the probes in time evolution of uv-vis spectra;fluorescent labeling to calculate oligonucleotides coverage of the probes. PCR-PAGE characterization for oligonucleotides of nanogold probes. Results After functionalized with antibody and thiol-capped oligonucleotides, the adsorbance peak of modified Au nanoparticles shifted from 518 to 524nm. Using gel electrophoresis we detemined the DNA on the gold nanoparticles, and the antibody activity were also fine. The surface coverage of nanogold between (10±3)nm is (92±20) oligonucleotides per nanogold.Conclusions The Preparation of the dual biomolecule -labeled nanogold probes is quiet easy and simple .With high stable combining of antibody and thiol-capped oligonucleotides,the nanogold probes could be promise for detection of ultramicro-protein.
