The Early Micropropagation of Uenishiwase Persimmon

The early stage micropropagation of Uenishiwase persimmon (Diospyros kaki Thunb, cv. Uenishiwase) was studied. Shoots were initiated from the dormant buds. Effects of hormones on the shoot percentage, numbers of shoots per explants and shoot height were investigated. The result showed that modified Murashige and Skoog (MS) medium supplemented with 2 mg.L-1 Zeatin (ZT) was the most suitable for micropropagation of Uenishiwase. Suplementing 5 mg.L-1 6-benzylaminopurine (BA) or Indolacetic acid (IAA) in the medium containing 1 mg.L-1 ZT significantly increased proliferation rate and average height of explants. Adding 0.1 mg.L-1 Naphthaleneacetic acid (NAA) to 5 mg.L-1BA medium improved the shoot percentage and proliferation too.
