Study on Rapid Identification of Genus and Species Relationship of Nosema Bombycis
In order to explore the parasite genus and its species relationship, two sets of primer V1F/530R and KAI01N/KAI02N were used to identify three strains microsporidia isolated from silkworm (Guangzhou), Hemerophila, atrilineta, and Antheraea pernyi G-M, respectively. The PCR results showed that there were only one band at the size about 400 by for three microsporidians, respectively, in the set of primer V1F/530R amplicon, but in the primers set of KAI01N/KAI02N amplication, there only produced a band at size up to 800 by for Nosema bombycis (Guangzhou strain) from silkworm and Microsporidium spp. from H. artilineta, respectively, and no amplicon for N. antheraeae. Furthermore, the amplicon from the primer of V1F/530R and KAI01N/KAI02N were cloned and sequenced, and were analyzed by using Blastn tool in NCBI. The results showed that N. bombycis (Guangzhou strain) was highly close to Microsporidium spp. and N. antheraeae was apparently different from N. bombycis (Guangzhou strain) within Nosema genus. This method was supplied a new approach for quickly estimating the relationship between various microsporidia.
