Preparation of Silk Fibroin Film Cross-linked by Carbodiimide (EDC)

Regenerated silk fibroin film was prepared by using 1-ethyl-3- (3-dimethylaminopropyl) carbodiimide hydrochloride (EDC) as cross-linking agent and N- hydroxysuccinimide (NHS), 2-morpholinoethanesulfonic acid (MES) as assistant agent. The fine structure, water resistance, tensile strength and elongation at break and Youngs modulus of crosslinked film were examined and analyzed. The results showed that there were more silk I in crosslinked film when 10-15% (relative to silk fibroin) EDC were added to silk fibroin solution and dried at 60℃. But the content of silk I decrease remarkably and the proportion of amorphous domains should correspondingly increase when the addition of EDC was more than 20%. The water solubility and Youngs modulus of crosslinked film decreased remarkably when 10-15% (relative to silk fibroin) EDC were added to silk fibroin solution, while the elongation at break increased. NHS and MES could strengthen the effect of crosslinking as assistant agent.
