Identification of S1 Protein Receptor of Hemagglutinating Encephalomyelitis Virus

To identify Hemagglutinating Encephalomyelitis Virus (HEV) receptor in pig kidney cell lines (PK-15) cytomembrane, the S1 protein of HEV were expressed in Pichia Pastoris and purified. Polyclonal antibody to HEV was prepared with immunizing rabbits by injecting the purified S1 protein. PK-15 cytomembrane proteins were extracted and transferred to nitrocellulose membrance. Virus overlay protein binding assay (VOPBA) was performed by using the recombinant S1 protein for identifing the receptor protein. The result showed that the 1797 bp S1 gene fragment was obtained by PCR amplification, and recombinated yeast expression plamid was constructed sucessfully. After the recombinated strains were induced and expressed, there was a molecular weight of 73kDa in medium supernatant. The result of VOPBA suggested that PK-15 cytomembrane proteins were bound with S1 protein of HEV, and one clear protein band of 90kDa was found.
