Amplification and Verification of the Nisin Promoter from Lactic Acid Bacteria Predominately Inhabited in Chinese Traditional Dairy Products

Lactic acid bacteria (LAB) are no doubt the most widely used probiotics with food grade status. Though China abounds in the resources of LAB, research on them is still at the primary stage, especially on bacteriocins produced by them, such as Nisin. Fueled up by the basic researches and commercially usage of LAB, the construction of new industrial strains with excellent properties, specifically employing the advanced biotechnology (such as Protein Engineering, Gene Engineering and Cell Engineering, and so on), not only confers food scientists good opportunity as well as big challenge, but also will fundamentally change the traditional applying method of LAB in China. A reliable expression system, like worldwide popular NICE (Nisin-Controlled gene Expression) using the Nisin promoter, is essential in trying to enhance the beneficial functions of LAB when employing them to express certain homo-or heterologous proteins. In this study, PCR method was first used to amplify the Nisin promoter fragment from lactic acid bacteria predominately inhabited in Chinese traditional dairy products, and then this sequenced fragment was inserted into promoter-probe vector pNZ273. Thereafter the function of the cloned fragment was checked by histochemical detection of the β- glucuronidase activity. The result indicated that the cloned fragment could induce expression of the reporter gusA gene in pNZ273, suggesting suitable usage as a Nisin promoter.
