Detection of Lactic Acid Bacteria against Streptococcus mutans and Purification of Its Bacteriocin
One strain designated KLDS 1.0432 of the isolated LABs was found to inhibit the growth of the indicator streptococcus mutans. Its supernatant fluid lost antimicrobial activity when treated by trypsin, pepsin and proteinase K, which indicated that the antimicrobial compound was protein or peptide. Analysis of the 16S rRNA gene sequence indicated that KLDS 1.0432 was a strain of Lactobacillus helveticus. The bacteriocin was purified by using the Hiload 26/60 Superdex 75 prep-grade chromatographic column and the AKTA purifier 100 system. Finally, Tricine- SDS-PAGE technology was used to estimate the molecular size of the bacteriocin and its size was found to be between 7823Da and 14400Da.
