Phosphoinositide 3-Kinase and p38 MAPK Are Involved in bFGF-Indueed Endothelial Cell Migration

Objective: Migration of endothelial Cells is a key event in angiogenesis, Meanwhile, basic fibroblast growth factor (bFGF) is a potent angiogenic molecule. It has demonstrated that bFGF can induce migration of endothelial cells. However, the cell signaling pathways involved in bFGF-induced endothelial cells migration have not been shown yet. This study aimed to evaluate the impact of PI3Kinhibitor, wortmannin and p38 MAPK inhibitors, SB203580 and ERK1/2 inhibitor, PD98059 on bFGF-induced endothelial cells migration. Methods: Endothelial cells were isloated from the human umbilical vein by the explanted method. After bFGF (6ng/ml) treatment, the migration of endothelial cells was assessed by wound healing assay. Wortmannin (100ng/mol), SB203580 (10umollml), and PD98059 (20umol/ml) were used to inhibit the effects of bFGF respectively. Results: The distance of wound healing of bFGF-induced group was3.0+0.2, and that of Wortmannin group,SB203580 group and PD98059 group was6.6+0.2, 5.27+0.115, and 3.2+0.2 respectively. The results showed that bFGF (6ng/ml) resulted in a marked increase in the migration compared with the control (p<0.05), which was inhibited by Wortmannin (p<0.05) and SB203580 (p<0.05), but not PD98059 (p> 0.05). Conclusion: bFGF induced migration of endothelial cells. Phosphatidylinositol (PI) 3-kinase pathway andp38 MAPK pathway, but not ERK1/2 MAPK pathway, are likely involved in bFGF-mediated signal transduction leading to endothelial cell migration as well as carrying an anti-angiogenetic ability.
