Study on the immobilization of papain supported by HBS lignin
The new idea of enzyme flexible immobilization was based on the bridge of a flexible and hydrophilic chain between the support and enzyme, so the rigidity and hydrophobic property of the support can be improved. If enzyme was immobilized on such a carrier with flexible chain, the immobilization can reduce the loss of enzyme activity and maintain the conformation of enzyme. In this paper, the papain was immobilized on the copolymer of HBS lignin grafted with flexible and hydrophilic polyacrylamide chain. The conditions of papain immobilization were investigated. The optimum conditions for immobilization were at 80℃, pH=8. 0 for 24h. The influences of polyacrylamide chain and the ratio of polyacrylamide grafted on the amount of immobilized papain were also discussed.
