Studies on Ganoderma Submerged Fermentation and Its Product Extraction Technique
This research screened excellent strain of Ganoderma (Lingzhi) for submerged fermentation and medium formula. Technique of submerged fermentation and product extraction were also studied. Shaking bottle cultivation was adopted to culture Ganoderma mycelia. Discuss the effect of different carbon source and nitrogen source on extraction rate mycelia and polysaccharide. The quantity of extracellular polysaccharide in fermentation liquid precipitated by ethanol was regarded as the criterion to value the strain and medium under the controlling fermentation conditions of temperature 28℃±1, rotation 180r/min and culture period 6d. The excellent strains were from Xingzhou, Jingda and Sanming. The crude polysaccharide was over 3.60g/L in these strains. The best carbon source was glucose, sucrose or maltose. The most suitable nitrogen source was silkworm pupa powder, peptone or other organic nitrogen. The medium for fermentation consisted of 2 % glucose, 0.2 % silkworm pupa powder or peptone, 0.15% KH<,2>PO<,4>, 0.05% MgSO<,4>·7H<,2>O. The technique was preliminarily determined the technique to produce Ganoderma industrially and extract polysaccharide.
