Purification and Identification of Glycopeptides from Ganoderma lucidum Fruit Bodies Cultivated with Grass and Wood Log

This research studied the amount and properties of glycopeptides of Ganoderma lucidum cultivated with wood log (GLPW) and grass (GLPG), and investigated the feasibility of substitution of grass for wood log in cultivate Ganoderma lucidum. Glycopeptides were extracted and isolated from fruit bodies of Ganoderma lucidum followed by dialysis. The component sugar and molecular weight distribution of the glycopeptides were determined by GC and HPLC. The structure of the glycopeptides was detected by IR, <1>H NMR and <13> C NMR. The molecular weight of the glycopeptides of GLPW and GLPG was Mw 5.13×10<5> and 5.85×10<5>,Mη 6.21× 10<5> and 6.96×10<5> respectively. The glycopeptides from both GLPW and GLPG were linked mainly through β-glycosidic bond and consisted of 16 amino acids as well as Rha, Xyl, Fru, Gal and Glc (There was small amount of Man in GLPW). The physical properties and chemical components of GLPW and GLPG were very similar (or have minimal differences). The glycopeptide extracting rate of GLPG was 2.8 times to GLPW.
