Application of Genechip in S-genotyping of Pear Cultivars

Pear is an important fruit tree with abundant varieties and rich genetic resources. It has large cultivated area in the world. Pear displays gametophtic self-incompatibility (GSI) and its self-incompatibility genotype (S-genotype) is useful for arrangement of pollination varieties in commercial orchard as well as breeding programs. Several approaches for S-genotyping of cultivars have been developed such as crossing experiments, PCR-RLFP system, DNA sequence analysis and cDNA cloning etc. In this paper, the advantages and disadvantages of these methods were analyzed, and a new genechip-based method was put forward. According to the structural features of pear S-RNase gene, some oligonucleotide probes were designed and successfully applied in assay of the S-genotypes of pear cultivars, which provides foundation for pear S-genotyping with the method of S-RNase genechip.
