DNA Hybridization Detection by X-ray Photoelectron Spectroscopy
To establish a convenient and economic method to determine hepatoma-specific alpha-fetoprotein(AFP-L3) for diagnosis of hepatocellular carcinoma(HCC). AFP-L3 from serum of HCC patient was separated by a mini-column Lens culinaris agglutinin(LCA)- affinity chromatography. The levels of serum total AFP and separated AFP-L3 were detected by radioimmunoassay(RIA). ALP-L3 from serum of HCC patients was eluted clearly on the LCA-sepharose gel mini-column with a solution containing alpha- methyl-D-mannoside. The total AFP in serum was separated into three peaks during the elution course. The AFP-L3 was apart of total AFP. A ratio of more than 15 percent for ALP-L3 to total AFP in serum was considered as a specific marker for HCC diagnosis with higher sensitivity(92.7%) and specificity(88.2%). The present data suggested that the assay in this study is more sensitive, repeatable, convenient. ALP-L3 is a useful marker for early diagnosis of HCC.
