Effect of Zinc Oxide Nanoparticles in Different Size under UV Irradiation on SMMC-7721 Cancer Cells

We demonstrated the first use of X-ray photoelectron spectroscopy (XPS), a relatively sensitive and less time-dependent technique to study the hybridization of DNA. The target DNA strand labeled with chlorine-containing Hexachloro-Fluorescein (HEX) was hybridized to DNA arrays with four different probes. Every probe dot of the arrays was detected with XPS. The XPS Cl2p peak area was found to decrease with the increase of mismatched base numbers in DNA probes. The XPS Cl2p peak area ratio for perfectly matched probe to one base mismatched probe, two bases mismatched probe and three bases mismatched probe accorded well with the corresponding fluorescent intensity ratio from fluorescent measurements, evidencing that XPS is a potential tool for the analysis of DNA hybridization.
