Gold nanoparticle-based detection of transcription factor NF-κB

Transcription factor and sequence specific DNA interaction plays important role in drug genome and transcription therapy. Gold nanoparticles show high sensitivity, stability and compatibility for biological molecules as electrochemical intercalators. There is no report of the detection of sequence specific DNA binding transcription factor by gold nanoparticle catalyzed-Ag enhancement at interface between electrodes and electrolyte solutions. Here unimolecular hairpin oligonucleotides were self-assembled onto Au electrode surface and elongation on solid phase was carried out to double strand oligonucleotides with transcription factor NF-κB binding sites. Gold nanoparticle-catalyzed Ag deposition was detected by anodic stripping voltammetry (ASV) for NF-κB binding. It was indicated that the method for sequence specific DNA binding protein detection showed pronounced specificity, sensitivity and gave highlight for transcription regulation, operator site and functional gene inspection.
