Improved Synthesis of 1-O-acetyl-2, 3, 5-tri-O-benzoyl-β-D- ribofuranose from Inosine

Medicines of nucleotides and nucleotide acids are very important in chemosynthesis of medication, and the quantity of this kind of medicines is rapidly increasing. 1-O-acetyl-2,3,5-tri -O-benzoyl-beta-D-ribofuranose is a key mediate for synthesizing medicines of nucleotides and nucleotide acids, which was synthesized by using inosine as the starting material through benzoylation and acetylization. Effects of the ratio of reactants, reaction time, and reaction temperature on the yield of the product were studied. The results showed that, the proper decrease in reaction temperature and decrease in dosage of pyridine were propitious for the increase of the yield of the product. The optimal reaction conditions were obtained as follows: 9.0 g (0.034 mol) inosine was dissolved in 40 mL pyridine, and then 15 mL (0.129 mol) benzoyl chloride was added, followed by stirring the solution at 10 ℃ for 15 h. The product was dissolved in a mixture of 40 mL glacial acetic acid and 5 mL (0.083 mol) acetic anhydride, and then 3 mL oil of vitriol was added, the solution was stirred at 10 ℃ for 17 h to get the final product. By means of the above process, the total yield of 1-O-acetyl-2,3,5-tri-O-benzoyl-beta-D -ribofuranose was summed up to 74.93% (HPLC purity: 98.0%). The structure of the product was identified by IR, 1HNMR and MS.
