Preparation and Comparison of Two Kinds of Streptavidin GoldMagnetic Nanoparticles

Two kinds of streptavidin GoldMagnetic nanoparticles (SA GoldMagnetic NPs) were prepared by physical adsorption and covalent binding, respectively. SA GoldMagnetic NPs prepared by covalent reaction (cvSA GoldMagnetic NPs) were treated with cysteamine firstly which had both thiol group and amino group as a covalent bifunctional crosslinker and glutaraldehyde sequentially, then streptavidin was successfully immobilized onto GoldMagnetic NPs surface by covalent interaction via the reaction between aldehyde group on GoldMagnetic NPs surface and streptavidin. For comparison, SA GoldMagnetic NPs by the methods of physical adsorption (paSA GoldMagnetic NPs) were also prepared. Coupling capacity for streptavidin and stability of these two kinds of SA GoldMagnetic NPs were evaluated. The paSA GoldMagnetic NPs possess high streptavidin binding capacity and simple preparation procedure. However, compared with the physical adsorption, the covalent immobilization efficiency of streptavidin onto GoldMagnetic NPs were more stable and would offer more opportunities toward using magnetic composite nanoparticles for biomedical purposes.
