Analysis of the neurogenic potential of multipotent skin-derived precursors
Human SKPs grew in suspension as neurospheres in the presence of the mitogens basic fibroblast growth factor (bFGF) and epidermal growth factor(EGF) without serum. Limited expansion of these neurosphere-like aggregates would restrict the study and application of SKPs. The purpose of this study was to discover a method for rapid expansion of single cells present in the adult human dermis with subsequent induction of neural phenotypes and analysis their gene expression profile. In this study, SKPs were cultured and expanded in the presence of the mitogens bFGF and EGF, and then by adherent expansion in serum, reverse transcription-polymerase chain reaction (RT-PCR) approaches was used to detect the gene expression profile of SKPs. The proliferation speed of these SKPs were significantly higher and large numbers of precursors were generated in short time. SKPs expressed nestin, p75NTR, fibronectin, vimentin, Slug and Snail and could be differentiate into cells expressing neural phenotypes by neuron and gliocyte derived signals.
