17b-estradiol and CpG-ODN Synergetically Activate Plasmacytoid Dendritic cells

Physiological gender differences in immune capabilities suggest that sex steroid hormones such as 17b-estradiol (E2) may be involved in the regulation of the immunocompetence. In this paper, plasmacytoid dendritic cells (pDC) from murine spleen were treated with E2 or/and CpG-ODN (CpG) for 24h or 72h. Then the viability, costimulatory molecules expression, cytokines secretion and stimulatory capacity were analyzed. Our results showed that E2 and CpG increased the viability of pDCs, and upregulated the expression of CD80, CD86 and CD40 on pDCs. Both E2 and CpG also increased IFN-a secretion and the stimulatory capacity of pDCs. These data suggested that E2 and CpG might synergetically influence the immune responses by changing the viability, maturation, stimulatory capacity and cytokine secretion of pDCs.
