Effect of Inhibition of DLC-1 Gene Expression by RNA Interference (RNAi) on Cell Proliferation and Migration in Colon Cancer LoVo Cell Line
DLC-1 (deleted in liver cancer 1) is a potential tumor suppressor gene, which is inactive in liver carcinogenesis. To observe the effect of DLC-1 gene expression on cell proliferation and migration in human colon cancer cell line, RNAi Lipo-recombinent of DLC-1 gene(pGCsiDLC)were constructed and transduced into LoVo cells which are positive for DLC-1 gene expression. The results have shown that RNAi recombinent could effectively inhibit the expression of DLC-1 gene in LoVo cells. Additionally, MTT also shown depressed DLC-1 gene expression resulted in inhibition of LoVo cell proliferation. Cell migration assay confirmed that knocking down DLC-1 gene expression was followed by promoting in LoVo cell migration. Our observations suggest that DLC-1 gene was associated with LoVo cell proliferation and migration. DLC-1 is a potential suppressor gene in colon cancer LoVo cell line and may play an important role in colon cancer mechanism.
