Expression of hGITR<aa27-165> Gene and Preparation, Detection of its Polyclonal Antibody

Human glucocorticoid-induced tumor necrosis factor receptor (hGITR) is a type I transmembrane protein with homology to TNF receptor superfamily members. Ligation of murine GITR can enhance immunity reactions via several signal pathways. However, so far the certain function of hGITR is not clear. In this study, we constructed pQE30-hGITRaa27-165 prokaryotic expression plasmid and then expressed hGITRaa27-165 fusion protein, subsequently, prepared polyclonal antibody (pAb) against the fusion protein. ELISA, Western blot and FCM were used to identify the titer and specificity of the pAb. The effect of anti-hGITRaa27-165 pAb on the proliferation of human CD4+T cells was investigated by 3H-TdR incorporation test.
