Generation of Antibiosis-free Mutant of Bacillus cereus B-02

The bacterial B-02, isolated from the soil growing tomato infected by Botrytis cinerea in Zibo,is an effective Bacillus cereus strain to control B.cinerea. To research on mechanism of B.cereus B-02 against B.cinerea, the antibiosis-free mutant is constructed using transposon tagging technique, and the results obtained in the present study lay the foundation for future research on antagonistic related genes. In this study,pTV1, temperature-sensitive suicide vector with Tn917, was introduced by electroporation into B.cereus B-02.17 transformed B.cereus B-02 strains were screened resistant against chloromycetin, erythromycin and lincomycin, moreover, this effect is highly reproducible and appears to be constant. By constant high temperature, the transposon Tn917 successfully inserted the genome of B.cereus B-02 and 1670 mutants were screened that were resistant to erythromycin and lincomycin but susceptible to chloromycetin. Among them, 1 antibiosis-free mutant was screened against B. cinerea.
