Induction of ADH5 mRNA expression by formaldehyde and association with ADH5 rs1155449 SNP
Formaldehyde is a carcinogen with highly toxic effect on organisms. ADH5 gene encodes the human formaldehyde dehydrogenase and is important to the detoxification of formaldehyde. The purpose of this work is to explore how the human ADH5 mRNA expression is influenced by formaldehyde exposure and the relationship between ADH5 rs1155449 SNP located in intron1 and ADH5 mRNA expression induced by formaldehyde. The Human immortalized B lymphocytes of Chinese Han population were established by EBV-transformed protocol in our work. The proliferation inhibition toxicity was evaluated by MTT assay. The SNP of rs1155449 was detected by DNA sequencing. The real-time reverse transcription-polymerase chain reaction (RT-PCR) approach was used to estimate levels of ADH5 mRNAs of different human immortalized lymphocytes. It showed the maximum no inhibition toxicity effect on cell proliferation of formaldehyde was round the concentration of 0.001%. The frequencies of ADH5 rs1155449 homozygous(AA),heterozygous(AC) and homozygote (CC) were 52.94%, 29.41%, 17.65%, respectively. The ADH5 mRNA expression could be up-regulated by formaldehyde at the dose as low as 0.0005% for 48h exposure. But it showed the polymorphism of rs1154409 located in ADH5 intron1 had no association with the expression of ADH5 mRNA induced by formaldehyde. The further study should be explored in the other gene function and other ADH5 SNP.
