Suppression of Growth and Invasion Phenotypes in Glioblastoma U251 Cells by the LRR Domain of LRRC4
LRRC4 is a candidated tumor suppressor gene which was isolated by position candidate cloning strategy and 5-RACE at the chromosome 7q31-32. Deduced amino acid sequence of LRRC4 by SMART software indicates that it contains an extracellular conserved LRR domain and an IgC2 domain. In the present study, the strategy of constructing deletion mutants (pcΔLRR, pcΔIgC2 or pcΔTm) of different domain by One-step PCR, was applied. These mutants were transfected into U251 cells, and U251 cell lines of stable expressed mutants were constructed by liposome transfection. We demonstrated that the full length LRRC4 could inhibit U251 cells proliferation and invasion in vitro by MTT, soft agar clony formation assay and transfilter cell invasion assay. However, mutant of LRR domain deletion couldnt inhibit U251 cells proliferation and invasion. Mutants of IgC2 or Tm deletion still inhibited U251 cells proliferation and invasion. Therefore, LRR domain is indispensable to inhibit U251 cells growth and invasion for LRRC4.
