Intranasal immunization with codon optimized MOMP DNA vaccine confers protection against Chlamydia trachomatis infection
Chlamydiae trachomatis is the leading cause of sexually transmitted bacterial disease worldwide. However there is currently no licensed vaccine against it. Our former research proved that humanized codon optimization can significantly enhance the expression of C. muridarum MOMP DNA. In this study we evaluated the immunological effect of codon modified MOMP(HuMOMP) DNA vaccine by intranasal immune route. Results showed that mice immunized with naked HuMOMP DNA vaccine were weak in inducing specific immune responses. When in combination with cholera toxin subunit B(CTB) and CpG as adjuvant, it produced higher levels of antigen-specific IgG in serum and IgA in vaginal fluid, showed stronger DTH reaction and proliferative T cell responses, produced higher IFN-gamma and therefore resulted in significantly enhanced clearance of C. muridarum intravaginal infection in mice than those without adjuvant. In conclusion, intranasal immunization with the codon optimized MOMP DNA vaccine confers protection against Chlamydia trachomatis genital infection.
