Pilot study on up-regulation of NGF-mRNA of mice induced by Formaldehyde

Nerve growth factor (NGF), together with brain-derived neurotrophic factor (BDNF) and and neurotrophins (NT)-3, NT-4/5, belongs to the neurotrophin family. NGF is synthesized by various structural and inflammatory cells, and it also can induce production, proliferation, differentiation and maturation of immuno-inflammatory cells, mediate inflammatory cells recruitment, and stimulate sensory neurons to secrete neuropeptide, especially substance P. Recently, some reports indicate that NGF play a key role in bronchial hyperresponsiveness, airway inflammation and bronchial remodeling, main features of asthma. And many researches show that formaldehyde (FA) exposure increased the risk of asthma. In order to explore whether FA can induce asthma by NGF way, this study takes mice mast cells, lung tissue of mice as materials, applies RTPCR to assess NGF mRNA expression levels in these rodent cells after FA exposure in vitro and vivo. The results showed that NGF mRNA expression in mice mast cells increased with FA concentration increasing, but it dropped sharply at 125 M FA concentration in vitro, and there was significant difference of the mice lung cells NGF mRNA expression levels between control groups and 3.0 mg/m3 groups for 1 day FA-inhaled in vivo (P<0.01). Consequently, FA can induce NGF-mRNA up-regulation of mice cells. This study shows us a possible mechanism that FA induced asthma by NGF pathway.
