MANIPULATION OF COOLING RATE DURING CRYOPRESERVATION OF HYPERICUM PERFORATUM L. SHOOT TIPS USING SLOW FREEZING METHOD

Shoot apices isolated from in vitro cultured Hypericum perforatum L. plants were subjected to cryopreservation using the slow freezing method. The plants regenerated from explants that survived cryopreservation procedure restored their developmental competence, genetic stability at both, cytogenetic (chromosome number, mitotic activity) and a DNA (VNTR sequences) levels and ability to synthesise bio-active substances hypericins. The viability of cryopreserved shoot tips was genotype-dependent and in average did not exceed 20 percent. Amongst the variables that contribute to the survival rate, manipulation with the cooling rates of the cryoprotective solution between the temperatures prior to phase transition and – 40 ℃ was performed. Experimental data were compared with computer simulation.
