HPCE Analysis of The Effect of Sargassum Fusiforme Polysaccharides on The Complex Mobility of Erythrocytes in Tumor-bearing Organisms

This study was to analyze the effect of Sargassum fusiforme polysaccharides (SFPS) on the electrophoretic mobility of erythrocytes in tumor-bearing organisms. A tumor mouse model was developed, and the mice were treated with high, medium, and low dosages of SFPS through abdominal injection for 7 d. Erythrocytes were collected and prepared into suspensions, and the complex motilities of the cells were measured using high performance capillary electrophoresis (HPCE). Experimental conditions: capillaries: 75 μm × 50 cm;buffer for electrophoresis: phosphate solution containing HPMC (0.1 mol/L, pH 7.4); injection pressure: 3.448 kPa; injection time: 10 s; separation voltage: 20 kV; column temperature: 25℃. The electroosmotic mobility was found to be 2.16×10-4 cm2·V-1·s-1, so SFPS can decrease the migration time of erythrocytes in tumor-bearing organisms, which was 17.56 for the control group, 13.96 for the low-dosage group, 12.9 for the medium-dosage group, 13.51 for the high-dosage group, while that for the normal group was 11.51; and increase its complex mobility, which was 1.06×10-4 cm2·V-1·s-1 for the control group, 1.19×10-4 cm2·V-1·s-1 for the low dosage group, 1.29×10-4 cm2·V-1·s-1 for the middle dosage group, 1.23×10-4 cm2·V-1·s-1 for the high dosage group, while that for the normal group was 1.45×10-4 cm2·V-1·s-1. The erythrocyte mobility for all three dosage groups differ significantly from that for the negative control (P<0.01). SFPS can change the complex mobility of erythrocytes from tumor-bearing organisms toward the level for erythrocytes from normal organisms, and this may be because it can change the density of the surface charge on erythrocytes. So HPCE can be used as auxiliary tool for determining the physiological state and functions of erythrocytes.
