IDENTIFICATION OF GENETIC POLYMORPHISMS IN THE MPO GENE USING ADAPTER-LIGATION MEDIATED ALLELE-SPECIFIC AMPLIFICATION

The difference of Myeloperoxidase (MPO) expression is related to many diseases including leukemia. MPO expression may be associated with the promoter polymorphisms and the extron polymorphisms, but the present studies were mainly focus on the -468G>A MPO promoter polymorphism. To further investigate the association of MPO gene polymorphisms with risk of leukemia in a Chinese population, we applied the adapter-ligation mediated allelespecific amplification (ALM-ASA) assay to determine five promoter polymorphisms and four extron polymorphisms in the MPO gene. An initial PCR amplified the relevant gene fragment containing all single-nucleotide polymorphisms (SNPs) of interest. This product was then digested by restriction enzyme and ligated with a DNA adapter. The adapter-ligated DNA was genotyped by allele-specific amplification. The association of MPO polymorphisms with leukemia was analyzed by a case-control study from 135 incident cases of leukemia and 187 controls. Five promoter polymorphisms ( - 1 816T>C, -1 468G>A, -1 053T>C,-5 86T>C,-468G>A) and four extron polymorphisms (53G>T, 90C>T, 93C>A, 115C>T) were analyzed. The typing results were consistent with those obtained by Sangers sequencing. Four variant alleles, - 1 816C,- 586C,- 468A and 53T, were detected and the analysis of these variants showed no association with leukemia, although a near significant decrease in -468A was found in leukemia group( 15. 9% vs 22.5%, P=0. 07). The ALM - ASA assay is simple, reliable, rapid, and amenable to automation and could facilitate the large-scale genotypic analysis of MPO. - 468A may be associated with a reduced risk of leukemia development.
