MOLECULAR CLONING PROPHENOLOXIDASE FROM THE CHINESE MITTEN CRAB ERIOCHEIR SINENSIS AND ITS EXPRESSION IN TREMOR DISEASE

The Chinese mitten crab Eriocheir sinensis is an important valuable freshwater aquicultural crustacean in China. But in the last few years, the aquiculture of crab has been particularly badly hit by epidemics of a serious disease, tremor disease. The prophenoloxidase (proPO) is a key protein in the innate immune system in crustacean and stored in haemocyte granules. The enzyme can be released into the hemolymph against infectious pathogen. But so far no information was reported on cDNA sequence of proPO in the crab and the relation of proPO mRNA expression to the infectious diseases with Spiroplasma. In this study, we cloned 202 bp cDNA sequence of proPO isolated from haemocytes of E. sinensis by a reverse-transcription polymerase chain reaction (RT-PCR). By comparison with other decapods, the 202 bp nucleic acid sequence of E. sinensis proPO exhibited the highest similarity to that of Cancer magister (87%), Scylla serrata (85%), and Padfastacus leniuscutus (84%). In addition, the crabs were infected by Spiroplasma, considered to be a pathogen to induce tremor disease of crab. Haemocytes were obtained from the infected crabs at 12 h, 24 h, 48 h, 72 h, 5 d, 10 d and 15 d respectively with 5 crabs in each time point. The mRNA expression of proPO was analyzed by RT-PCR, The results showed that the low mRNA expression of proPO was detected in control crabs. The expression levels of proPO significantly changed with infectious course of Spiroplasma and symptom of tremor disease. This suggests that the proPO expression is related to the health status of crabs and the proPO system in E. sinensis may play an important role in non-self recognition and host immune reactions against Spiroplasma.
